Chinese Scientist Apologises For "Leak" Of Baby Gene-Editing Result

Hong Kong, Nov 28: The Chinese scientist who claims to have created the world's first genetically-edited babies said Wednesday the trial had been paused, following an international outcry over the highly controversial procedure.

He Jiankui defended his work in front of a packed Hong Kong biomedical conference, saying he had successfully altered the DNA of twin girls born to an HIV-positive father, an apparent medical first.

A total of eight volunteer couples -- HIV-positive fathers and HIV-negative mothers -- had signed up to the trial, with one dropping out before it was halted.

He said there had been "another potential pregnancy" involving a second couple, but when questioned further agreed it had been a chemical pregnancy -- a term referring to a very early miscarriage.

"I must apologise this result was leaked unexpectedly," He said of the apparent breakthrough "The clinical trial was paused due to the current situation," he added.

The conference has been upended by the gene-edited baby revelations claims, after university professor He posted a video claiming the twin girls -- born a few weeks ago -- had had their DNA altered to prevent them from contracting HIV.

The move prompted a heated debate among the scientific community, with many raising concerns over the lack of verified data and the risks of exposing healthy embryos to gene editing.

Editing human embryos can create unintended mutations in other areas -- so-called "off-target effects" -- which can be carried through to birth, experts warned.

But He took to the stage Wednesday to justify his work, and was bombarded with questions as he told the audience that the parents were aware of the potential dangers when they signed up.

"The volunteers were informed of the risk posed by the existence of one potential off-target and they decided to implant," he said.

He also said the university where he works had been "unaware of the study's conduct."

Southern University of Science and Technology, in the southern Chinese city of Shenzhen, had earlier distanced itself from He, saying he had been on unpaid leave since February and had "seriously violated academic ethics and codes of conduct".

Organisers of the Second International Summit on Human Genome Editing, which opened Tuesday, also said they had been unaware of He's work.

Conference moderator Robin Lovell-Badge said He's trial was a "backward step" for the science industry, but described the babies' birth as "momentous" nonetheless.

"This is an example of an approach that was not sufficiently careful and cautious and proportionate," he said.

"Clearly however it is a point in history... These two babies would appear to be the first gene-edited babies. So it is a momentous point in history."

Summit chair David Baltimore, a Nobel laureate, said there had been "a failure of self-regulation by the scientific community because of a lack of transparency."

He's claim would "be considered irresponsible", Baltimore said.

Bypassed ethical process

He, who was educated at Stanford University, said the twins' DNA was modified using CRISPR, a technique which allows scientists to remove and replace a strand with pinpoint precision.

Gene editing is a potential fix for heritable diseases but it is extremely controversial because the changes would be passed down to future generations and could eventually affect the entire gene pool.

In many countries the editing of human DNA is tightly controlled.

Qiu Renzong, formerly the vice president of the Chinese Ministry of Health's ethics committee, told reporters at the conference that lax regulations in China mean that scientists who break the rules often face no punishment, and think of the ministry as being "without teeth".

China's National Health Commission ordered an "immediate investigation" into the case, the official Xinhua news agency reported, while the Shenzhen hospital meant to have approved the research programme denied its involvement.

A union of Chinese scientists issued a statement saying it "resolutely opposes so-called scientific researches and biotech applications that violate the spirit of science and ethics," Xinhua said.

The case has damaged China's international reputation in the field, said the Chinese Union of Life Science Societies.

This is not the first time Chinese researchers have experimented with human embryo technology.

Last September, scientists at Sun Yat-sen University used an adapted version of gene-editing to correct a disease-causing mutation in human embryos.

There is also a history of fraud within China's academic community -- including a scandal last year that led to the withdrawal of 100 "compromised" academic papers.

A joint statement Monday from a group of 100 scientists in China criticised He Jiankui's claims and called them a "great blow to the global reputation and development of biomedical research in China".

Washington DC, Jun 29: Young children with narrow retinal artery diameters were more likely to develop higher blood pressure, and children with higher blood pressure levels were more likely to develop retinal microvascular impairment during early childhood, according to a new study.

The first study to show this connection in children was published today in Hypertension, an American Heart Association journal.

High blood pressure, the main risk factor for the development of cardiovascular disease (CVD), can manifest as early as childhood, and the prevalence of high blood pressure among children continues to rise. In previous studies, analysis of blood vessels in the retina has shown promise as a predictor of CVD risk among adults. In the study titled, "Retinal Vessel Diameters and Blood Pressure Progression in Children," researchers sought to predict the development of high blood pressure in children over four years based on retinal blood vessel measurements.

"Hypertension continues as the main risk factor for the development of cardiovascular diseases and mortality," says Henner Hanssen, M.D., the study's lead author and a professor in the department of sport, exercise and health at the University of Basel in Switzerland. 

"Primary prevention strategies are needed to focus on screening retinal microvascular health and blood pressure in young children in order to identify those at increased risk of developing hypertension. The earlier we can provide treatment and implement lifestyle changes to reduce hypertension, the greater the benefit for these children."

Researchers screened 262 children ages six to eight from 26 schools in Basel, Switzerland, in 2014, for baseline blood pressure and retinal arterial measurements. Both measures were taken again in 2018. Blood pressure measurements at both baseline and follow-up were performed in a sitting position after a minimum of five minutes of rest and were categorized based on the American Academy of Pediatrics' blood pressure guidelines. These guidelines utilize the same measurements as the American Heart Association/American College of Cardiology 2017 Guideline for the Prevention, Detection, Evaluation, and Management of High Blood Pressure in Adults.

Results from the analysis indicate: children with narrower retinal vessel diameters at baseline developed higher systolic blood pressure at follow-up; retinal vessel diameters could explain 29 -31 per cent of the changes in systolic blood pressure progression between 2014 and 2018; children with higher blood pressure levels at baseline developed significantly narrower arteriolar diameters at follow-up, depending on weight and cardiorespiratory fitness; and initial blood pressure measures explained 66-69 per cent of the change in retinal arteriolar diameter from baseline to follow-up.

"Early childhood assessments of retinal microvascular health and blood pressure monitoring can improve cardiovascular risk classification. Timely primary prevention strategies for children at risk of developing hypertension could potentially counteract its growing burden among both children and adults," said Hanssen.

Researchers noted limitations of their study include that they could not confirm blood pressure measurements over a single 24-hour period, so they would not account for "white coat" hypertension, a condition where patients have high blood pressure readings when measured in a medical setting.

Developmental stage including puberty status of each child was not accounted for in the study, as well as genetic factors or birth weight - variables that could impact blood pressure development and microvascular health.

In addition, reference values for appropriate retinal vessel diameters in children do not currently exist, so future studies are needed to determine age-related normal values during childhood.

Chandigarh, Jan 15: A man, who has killed two women over infidelity over the last 10 years was arrested from a news channel studio in Chandigarh when he confessed to these crimes during a live programme, police said.

In the television programme on News18, the 31-year-old accused, Maninder Singh, who is a cab driver, confessed killing his live-in partner, a 27-year-old nurse Sarabjit Kaur at a Chandigarh hotel on New Year's Eve.

Singh, a former murder convict and currently out on bail, also confessed about his crime committed in Karnal in 2010.

"I killed her (Sarabjit Kaur) because she was having an affair with her sister-in-law's brother," Maninder told the news channel.

Confessing his previous crime, Maninder said he had killed Renu in Karnal. "She was also having an affair with a man from Uttar Pradesh," he said.

Singh was arrested while the programme was still on air as police rushed into the studio.

Haryana Police had arrested him for killing the woman in 2010. He was convicted by a trial court, but he later got bail from the Punjab and Haryana High Court.

London/Milan, Jun 2: World Health Organization experts and a range of other scientists said on Monday there was no evidence to support an assertion by a high profile Italian doctor that the coronavirus causing the COVID-19 pandemic has been losing potency.

Professor Alberto Zangrillo, head of intensive care at Italy's San Raffaele Hospital in Lombardy, which bore the brunt of Italy's COVID-19 epidemic, on Sunday told state television that the new coronavirus "clinically no longer exists".

But WHO epidemiologist Maria Van Kerkhove, as well as several other experts on viruses and infectious diseases, said Zangrillo's comments were not supported by scientific evidence.

There is no data to show the new coronavirus is changing significantly, either in its form of transmission or in the severity of the disease it causes, they said.

"In terms of transmissibility, that has not changed, in terms of severity, that has not changed," Van Kerkhove told reporters.

It is not unusual for viruses to mutate and adapt as they spread, and the debate on Monday highlights how scientists are monitoring and tracking the new virus. The COVID-19 pandemic has so far killed more than 370,000 people and infected more than 6 million.

Martin Hibberd, a professor of emerging infectious disease at the London School of Hygiene & Tropical Medicine, said major studies looking at genetic changes in the SARS-CoV-2 virus that causes COVID-19 did not support the idea that it was becoming less potent, or weakening in any way.

"With data from more than 35,000 whole virus genomes, there is currently no evidence that there is any significant difference relating to severity," he said in an emailed comment.

Zangrillo, well known in Italy as the personal doctor of former Prime Minister Silvio Berlusconi, said his comments were backed up by a study conducted by a fellow scientist, Massimo Clementi, which Zangrillo said would be published next week.

Zangrillo told Reuters: "We have never said that the virus has changed, we said that the interaction between the virus and the host has definitely changed."

He said this could be due either to different characteristics of the virus, which he said they had not yet identified, or different characteristics in those infected.

The study by Clementi, who is director of the microbiology and virology laboratory of San Raffaele, compared virus samples from COVID-19 patients at the Milan-based hospital in March with samples from patients with the disease in May.

"The result was unambiguous: an extremely significant difference between the viral load of patients admitted in March compared to" those admitted last month, Zangrillo said.

Oscar MacLean, an expert at the University of Glasgow's Centre for Virus Research, said suggestions that the virus was weakening were "not supported by anything in the scientific literature and also seem fairly implausible on genetic grounds."
