More reasons to get a goodnight's rest. Sleep-deprived brain can't form memories

Washington, Feb 13: If you don't get enough sleep, you might just start forgetting things. Sleep deprivation can interfere with the process that helps reinforce our memories, a new study has found.

sleepAfter studying mice, researchers at Johns Hopkins University in the US have fortified evidence that a key purpose of sleep is to recalibrate the brain cells responsible for learning and memory so the animals can ‘solidify' lessons learned and use them when they awaken.

The researchers also report they have discovered several important molecules that govern the recalibration process, as well as evidence that sleep deprivation, sleep disorders and sleeping pills can interfere with the process.

“Our findings solidly advance the idea that the mouse and presumably the human brain can only store so much information before it needs to recalibrate,” said Graham Diering from Johns Hopkins University.

“Without sleep and the recalibration that goes on during sleep, memories are in danger of being lost,” said Diering.

Diering said that current scientific understanding of learning suggests that information is “contained” in synapses, the connections among neurons through which they communicate.

On the “sending side” of a synapse, signalling molecules called neurotransmitters are released by a brain cell as it “fires”; on the “receiving side,” those molecules are captured by receptor proteins, which pass the “message” along.

If a cell receives enough input through its synapses, it fires off its own neurotransmitters.

More specifically, experiments in animals have shown that the synapses on the receiving neuron can be toggled by adding or removing receptor proteins, thereby strengthening or weakening them and allowing the receiving neuron to receive more or less input from nearby signalling neurons.

When neurons are “maxed out” and constantly firing, they lose their capacity to convey information, stymying learning and memory. One possible reason that neurons do not usually max out is a process that has been well-studied in lab-grown neurons but not in living animals, asleep or awake.

Known as homeostatic scaling down, it is a process that uniformly weakens synapses in a neural network by a small percentage, leaving their relative strengths intact and allowing learning and memory formation to continue.

To find out if the process does occur in sleeping mammals, Diering focused on the areas of the mouse brain responsible for learning and memory: the hippocampus and the cortex.

He purified proteins from receiving synapses in sleeping and awake mice, looking for the same changes seen in lab-grown cells during scaling down.

Results showed a 20% drop in receptor protein levels in sleeping mice, indicating an overall weakening of their synapses, compared to mice that were awake.

The study appears in the journal Science.

Washington, May 19: As the scientists across the world are struggling to develop a vaccine for combating coronavirus, US drugmaker Moderna announced on Monday (local time) that the phase I trial of its Covid-19 vaccine has shown positive early results.

The company is hopeful that it's vaccine could be available to the public as early as January next year. Several firms across the world are in the race to develop a vaccine for the deadly virus which has claimed over 3 lakh lives worldwide.

CNN citing Dr. Tal Zaks, Moderna's chief medical officer reported that "if future studies go well, the company's vaccine could be available to the public as early as January".

"This is absolutely good news and news that we think many have been waiting for for quite some time," Zaks was quoted as saying.

Moderna, based in Cambridge, Massachusetts announced that the vaccine developed neutralising antibodies to the virus at levels reaching or exceeding the levels seen in people who have naturally recovered from Covid-19, reported CNN.

These will be followed by phase 2 trials and phase 3 trials, which Moderna plans to start in July.

President Donald Trump had on Friday said that that the United States will be able to deliver a few hundred million doses of COVID-19 vaccine, under 'Operation Warp Speed', by the end of this year.

"I have very recently seen early data from a clinical trial with a coronavirus vaccine and this data made me feel even more confident that we'll be able to deliver a few hundred million doses of vaccine by the end of 2020 and we will do the best we can," Trump had said at a press conference at the White House on Friday.

Washington, May 21: US President Donald Trump China is on a "massive disinformation" campaign and is desperately trying to deflect the "pain and carnage" that it spread throughout the world, US President Donald Trump has said, upping the ante on Beijing over its handling of the coronavirus outbreak.

Trump, who has expressed disappointment over China's handling of the COVID-19 pandemic, claimed that it was the "incompetence" of Beijing that led to the mass killing across the globe. 

"China is on a massive disinformation campaign because they are desperate to have Sleepy Joe Biden win the presidential race so they can continue to rip-off the United States, as they have done for decades, until I came along!" Trump said in a tweet on Wednesday.

"Spokesman speaks stupidly on behalf of China, trying desperately to deflect the pain and carnage that their country spread throughout the world. Its disinformation and propaganda attack on the United States and Europe is a disgrace… It all comes from the top. They could have easily stopped the plague, but they didn't," he said in a series of tweets.

Trump blamed China for spreading the coronavirus globally and accused it of being incompetent.
"Some wacko in China just released a statement blaming everybody other than China for the Virus which has now killed hundreds of thousands of people. Please explain to this dope that it was the 'incompetence of China', and nothing else, that did this mass Worldwide killing!" Trump said.

China has denied covering up the extent of its coronavirus outbreak and accused the US of attempting to divert public attention by insinuating that the virus originated from a virology laboratory in Wuhan.

"China was the first country to report the COVID-19 to the World Health Organisation (WHO), (and) that doesn't mean the virus originated from Wuhan... There has never been any concealment, and we'll never allow any concealment," Chinese Foreign Ministry spokesman Zhao Lijian said last month.

"A discerning person will understand at a glance that the purpose is to create confusion, divert public attention, and shirk their responsibility," he said.

The novel coronavirus which first originated in Wuhan in December last year has claimed 328,120 lives and infected nearly 5 million people globally. The Us is the worst affected country with 93,439 deaths and over 1.5 million infections, according to Johns Hopkins University data.

Meanwhile, the US Senate passed a bill boosting oversight of companies based in China and other nations that could lead to their removal from American stock exchanges.

The Holding Foreign Companies Accountable Act, proposes to increase oversight of Chinese and other foreign companies listed on American exchanges and delist and ban over-the-counter trading for firms that are out of compliance with US regulators for a period of three years.

In a related development, a group of top Republican Senators led by Marco Rubio sent a letter to Secretary of the Treasury Steven Mnuchin following disturbing reports that China's state-owned and-directed enterprises were looking to exploit the economic crisis by buying US and foreign companies.

As companies backed by the Chinese Communist Party (CCP) approach banks to identify the purchase of companies in the US and in Europe affected by the pandemic, the senators urged Mnuchin to protect against the China's and the CCP's predatory economic behaviour during the COVID-19 crisis.

"We write to express our concerns related to the People's Republic of China's (PRC) efforts to exploit the economic crisis wrought by the COVID-19 pandemic to gain control of distressed companies or shirking its international responsibilities amidst a worldwide crisis.

"In both Chinese Communist Party (CCP) and PRC policy documents, Beijing has made no secret of its intentions to dominate strategic industrial and emerging technology sectors as well as influence standards at the expense of liberal, rules-based governance," wrote the senators.

As the crisis reverberates across the globe, the PRC's predatory lending practices — including the use of non-disclosure agreements for bilateral loans — not only damage the fiscal situation of recipient countries but also undermine the international community's ability to respond effectively to the crisis, they said.

"Without US and international pressure for accountability and transparency, those countries that are in debt to the PRC will not have the political cover or protection to open their financial books. Such countries will face the risk of default or a currency crisis, leaving the International Monetary Fund (IMF), World Bank, and Western countries to clean up the PRC's mess," the senators said.

During a campaign round table Katrina Pierson, Senior Advisor to the Trump 2020 Campaign, said that only the US President will defeat the coronavirus, hold China accountable for their negligence, and defend the American people from socialism.

If everything goes as Russia’s expectation, it will be world’s first country to approve a coronavirus vaccine for widespread in the second week of August despite safety and efficacy concerns, according to a report. The adenoviral vector-based vaccine developed by Russian military and government researchers is currently in phase 2 trials.

According to a report from CNN, Russian officials are hoping to get approval for the COVID-19 vaccine developed by the Moscow-based Gamaleya Institute on Agust 10 or even before that. The officials told the outlet that the vaccine will be approved for public use with frontline healthcare workers receiving it first.

“It’s a Sputnik moment,” said Kirill Dmitriev, head of Russia’s sovereign wealth fund, which is financing Russian vaccine research, referring to the successful 1957 launch of the world’s first satellite by the Soviet Union, according to CNN.

“Americans were surprised when they heard Sputnik’s beeping. It’s the same with this vaccine. Russia will have got there first,” he was quoted as saying.

However, Russia is yet to release the scientific data on its coronavirus vaccine trials, hence, questions remain about the safety and efficacy of the vaccine, said the report. The vaccine is in the second phase of testing with developers planning to launch the phase 3 trials sometime after August 3.

Earlier, Interfax reported, citing Health Minister Mikhail Murashko, that the vaccine will be widely used in parallel with phase 3 trials. He said the country plans to vaccinate medics who are at high-risk for COVID-19 next month before clinical trials are completed.

Murashko added that individuals at higher risk of getting infected with the coronavirus such as older people or those with health conditions will also be prioritised for the vaccine, although he did not estimate or reveal when that would happen. The minister added 800 people will be recruited for the phase 3 trials.

Meanwhile, health officials were more cautious considering the fact that human testing of the vaccine is incomplete, and the state registration is expected to begin after August 3, reported The Moscow Times.

On Monday, Moderna and Pfizer announced the commencement of the final phase 3 trials of their candidate vaccines against the SARS-CoV-2 virus, which has so far claimed at least 654,477 lives and infected 16,514,500 people worldwide. 

Researchers will recruit up to 30,000 volunteers in separate trials both backed by the US government. India's first indigenous coronavirus vaccine, COVAXIN, is undergoing phase 1 human clinical trials across the country. More than 150 COVID-19 vaccines are being developed all over the world with at least six candidates already in late-stage clinical trials.
